Nanobodies are the smallest antigen-binding fragments of heavy chain-only antibodies, with a molecular weight of 12–15 kDa. Nanobodies allow a broad range of applications in biotechnology, diagnosis, and therapeutics due to their small size, stability, simple production, high affinity as well as the benefit of multiple reformatting opportunities.
Nanobodies comprise naturally occurring variable domains derived from camelids and sharks as well as synthetic combinatorial libraries. All custom nanobodies will be derived from our fully synthetic phage/yeast display libraries. The high quality of the libraries and optimized protocols ensure a high success rate of isolating specific nanobodies against virtually any protein target. We have successfully isolated high-affinity nanobodies against multiple targets including transmembrane proteins and peptides.
The library design was based on the camelid VHH. Its framework was humanized, and the amino acids in CDR1-3 were fully randomized and synthesized with trimers. The theoretical diversity (DNA) of the library is more than 10^20, and the actual diversity (cell clones) is about 10^9.
